100-Fold Improvement in Detection Limit of Group-A Strep Lateral Flow Immunoassays using Isotachophoresis
We present a rapid, easy to use point-of-care immunoassay diagnostic for the detection of group-A streptococcus (GAS) with a limit of detection that is two-orders of magnitude lower than that of a commercial lateral flow assay. The limit of detection is dramatically improved by applying a tiny electrical current and performing isotachophoresis, a powerful electrokinetic technique to concentrate target proteins and enhance signal in lateral flow assays. The strategy we employ is applicable to variety of diseases where proteins are a potential target of interest. This dramatic improvement in analytical sensitivity may enable the use of lateral flow based rapid diagnostics in a wide range of point-of-care diagnostic applications.
